Melt Curve Analysis for Isothermal Amplification

Axxin instruments can facilitate melt curve acquisition and analysis for isothermal assays.
Nucleic acid (NA) amplification is often required to detect small quantities of DNA or RNA in molecular diagnostics. Once amplified, the specificity of the resulting amplicon and the presence of primer-dimers or non-specific amplicons can be identified using techniques such as gel electrophoresis, however, this process can be time consuming and relies on additional equipment and preparation. Alternatively, melt curve analysis can be conducted in the same tube the sample has been amplified in, potentially running automatically after the amplification step, saving time and resources while providing confidence in the product specificity.
Axxin has developed accurate melt curve analysis using continuous temperature ramping, rather than stepped temperature increments, optimised for isothermal assays. Melt curve analysis is available on the Axxin Release 3 (R3) T8-ISO Instrument software updated to v6 or later. It is also included in new instruments that ship with v6 or later instrument software.

The melt curve function is configured as part of the test type and runs automatically at the end of amplification (unless additional dyes are required prior to performing melt curve). The melt curve function is integrated within new Desktop Software for setup and analysis.
